Zusammenfassung NMR on hyperpolarized 129Xe (HpXe) offers tremendous potential to yield biomedical spectroscopic and imaging information based on molecule-specific signals. We have introduced a type of 129Xe-biosensor which enables the detection of immunological relevant complex formation of a peptide ligand with MHC class-II protein. To perform MR experiments with optimal sensitivity a knowledge of HpXe exchange- and relaxation-rates is required. The relaxation time of bulk 129Xe and the applied flip angle can be determined by repetitive measurements of the 194 ppm amplitude varying TR between consecutive excitations. The exchange rate of caged xenon can be determined by a pulse train of three excitations (centered on 60 ppm). The first pulse is used to acquire the reference signal amplitude A0, the second to destroy/invert the polarization and the third excitation after a delay time tdel to determine the signal build up Aup/A0 due to exchange with bulk HpXe. By varying the sample temperature from 24°C to 35°C one can double the exchange rate from ˜50 s-1 to ˜100 s-1. The exchange- and relaxation-rates can be used to optimize hyper-CEST experiments which may then allow for sensing events in the 10 nM range in future applications.
